I tend to allow new batteries to run down completely before charging for the first few cycles and also do this periodically throughout the battery's life.
In fact, according to most manufacturers, this is completely unneccessary with modern batteries but I guess it can't hurt.
To get the most out of your battery, you're probably better doing all the standard battery saving things like:
- reduce screen brightness
- reduce the time before the screen times out
- turn off / reduce frequency for sync on high traffic apps like Facebook and Twitter
- turn off radios that are not in use
The Note has a large screen so the screen stuff may be help, but in general, these don't tend to make a life altering difference, unfortunately.
Your best bet is probably to keep any eye on what's using battery (you can see this in Settings .. Battery). Some apps can be battery heavy or just poorly coded. When you identify one, kill the process and either remove the app or use with caution in future.
If you're a really heavy user, get a spare battery or a battery pack (an external battery that charges your phone's battery).
