Help location tracking

Hi, I have a rooted phone running an app with no root permission, but permission to determine my precise location via GPS. Now, I have "Location" turned off in the android settings. Is it possible for this app to determine my location via GPS? Or do I need to have "Location" enabled manually? I am running Lollipop.
By the way, I know there are network-based and radio-based means of determining location; I am just asking about GPS.

If you are connected to a wifi network, your IP address can give a relative position. Not extremely accurate, but usually the city you are in at least.

I only tether and use TOR + VPN + Proxy. Wi-fi is always disabled. I know apps can turn it on if they have the permission, but they can't do much besides see a router's MAC address and SSID. My home IP is safe.
