To darken the screen on my phone (LG G3) I simply tap twice anywhere on the screen. However, out of habit, I always tap in the same spot. I've been doing this for 2 years now and wonder if this might ultimately damage the pixels in that area. Obviously easy enough to fix - just tap different places, but I was curious. Thanks.

No you won't damage anything on the screen by tapping on it. Unless you're Godzilla.

Your G3 has what's called an 'IPS LCD' (In-Plane Switching Liquid Crystal Display). You should be fine tapping it in that same spot on your screen, the odds are its back light will burn out just as a matter of aging and render your entire screen useless well before your muscle memory induced tapping will have an effect.
