Smart actions will do it.
As an example, I have my phone set to automatically set to go to silent at x:xx pm and out of silent at y:yy am. Except that the phone will ring when my wife calls from her cell phone or work.
It looks like you should be able to set it up so calls from ABC and DEF always ring if you use VIP Caller Mode.

On a side note, make sure you temporarily disable this smart action when in a movie. Apparently, nobody else in the Dark Knight Rises cared that my alarm company called me, but they all found out anyways.
FWIW, you can limit that action using location (like your home) as a trigger.
I also set up a separate smart action that silences the ringer (vibrate) based on location as a trigger. You can specify the movie theater as a location. I use it for client locations, meeting and seminar venues, doctors offices, theaters, etc.
