I have a Samsung Galaxy Tab 2 7". When I'm reading a book I downloaded thru Nook, the sleep mode won't turn off the tablet when its idle.

Most book applications disable sleep mode when you have a book open to read. If you want the system to power off when its idle, you'll need to exit out of the nook app.
You can also check the Nook settings, it may have a "idle after..." setting or it may allow you to override the "do not allow to sleep" command.
