My boss lost his phone and I am busy typing them into excel. When he gets his replacement Samsung Galaxy note, will I be able to transfer them directly from the excel spreadsheet to his phone?

Why not open a new google account on a PC and type them into the contacts in that account. He can then add the account to his phone (as a second account) and google will sync them to his phone.
Have you checked that they were not google account contacts before, on his old phone. If they were they will be synced as soon as he registers his orignal google account on the replacement phone.
