Everything on the whole phone at once? Something yet to be written, unless you're talking about an iPhone. Android doesn't do it yet.
If you're talking about apps, data, photos, music, etc., Titanium Backup for apps and their data, and drag and drop (manually) or some PC backup program (like Karen's Replicator) for media.

I would recommend Helium. Its developed by ClockworkMod, and I personally use it. Its pretty great

I was looking at Helium, which is recommended by others as well. What stopped me was this line from the developer: "Note: Helium is NOT available to Motorola devices. This is because Motorola phones have a bug that break Android's backup mechanism. Until this is fixed by Motorola, Helium will not work on those Androids."
Does it work correctly and with the desktop app? Can I backup to my computer?
