Honestly, don't stress and just start playing. I was new to Android too when I got my Hero a month ago. Maybe 3 days before I felt fully comfortable.
Problem is. 100 people responding gonna net you 100 different responses. Just the nature of Android, there is no one way. Just your way. You will find yours soon enough.

Personally I would update the PRL and profile (both under settings, updates). I didn't know that and had horrible sound at first. As soon as someone on this forum told me about that I did it and all was well.
Also read some of the stickies about battery life. Those threads have some good lists about all the settings to tinker with to get the most out of the battery. Good luck and enjoy your new phone!
