To get the most benefit from either (though I've never used CPU tuner) you'll want to use an overclock kernel on the phone. Once a compatible kernel is used, you can then set the phone processor's maximum clock speed higher than its stock default value of 600MHz.
Make certain any "Set on boot" options are unchecked! If it is checked and you set too high a clock rate, you'll find yourself in a world of pain when your phone begins helplessly boot-looping.
