How does it work when I download profiles from the Tasker wiki? Do I just download them and activate them from the app, or will there be further configuration?

Using pre-made Tasker profiles

Download the profiles, then in Tasker use Menu/Profile Data/Import... to make them available. You can then edit them if required.
One thing I would point out is that Application contexts are tied to the package name, which can change from one phone model or manufacturer to another, even when the app name appears to be the same. So if you have problems with profiles not working when they use an Application context, you may need to edit the context and reselect the application.
